Zwf encodes glucose-6-phosphate dehydrogenase, which catalyzes the first step of the pentose phosphate pathway (PPP). This pathway supplies both biosynthetic precursors and reducing power in the form of NADPH, essential for anabolic metabolism. The reaction catalyzed by Zwf is frequently the rate-limiting step of the PPP and therefore represents a common target for overexpression in metabolic engineering [276]. Deletion of zwf effectively disrupts the PPP [322].
