The phosphopantetheinyl transferase (PPTase) sfp from Bacillus subtilis is required to activate the apo form of carboxylic acid reductase (CAR) [331]. Sfp catalyzes the transfer of a 4'-phosphopantetheine moiety from coenzyme A to a conserved serine residue in the peptidyl carrier protein (PCP) domain of CAR. This modification converts the inactive apo-CAR into the active holo-CAR, enabling it to form thioester intermediates necessary for catalysis.
